趋化因子是一类分子量小的细胞因子,其主要作用是在稳态和病理条件下募集白细胞亚群。趋化因子受体表达于细胞表面,是与G蛋白偶联的7次跨膜蛋白,趋化因子就是与受体结合后传递细胞信号的。通过趋化因子受体激活G蛋白后,紧接着激活磷脂酶C(PLC)。 PLC催化磷脂酰肌醇(4,5)-二磷酸(PIP2)分解生成肌醇三磷酸(IP3)和甘油二酯(DAG),两者都是第二信使分子,可以进一步扩大细胞内信号传导; DAG激活蛋白激酶C(PKC),IP3触发细胞内储存钙的释放。二者的激活可以触发许多信号级联反应(例如MAP激酶途径),如趋化性,脱粒,超氧化物阴离子的释放以及细胞粘附分子亲和力的变化。趋化因子也是通过这种机制来诱导响应细胞定向趋化。
